Enzymatic synthesis of 2'-deoxy[2,8-3H]adenosine and [2,8-3H]adenosine of high molar activity

Description
2'-Deoxy [2,8-3H]adenosine and [2,8-3H]adenosine were prepared by enzymatic synthesis catalyzed by the unpurified supernatant fraction of E. coli B homogenate. Both enzymes required for the catalytical transfer of the deoxyribosyl (or ribosyl) group were present in the unpurified fraction. The use of Trisbuffer as well as the presence of Mg2+ ions were found to be unnecessaryy for the reaction under study. Degrees of conversion exceeding 80 per cent were achieved. The reaction also proceeds at lower temperatures (297 and 278 K). 2'-Deoxy[2,8-3H]adenosine and [2,8-3H]-adenosine were prepared at molar activities of 1.68 TBq mmol-1 and 1.7 TBq mmol-1, respectively.
