Establishment of an RIA of serum thyroid peroxidase antibody and evaluation of its clinical use

Objective: RIA of serum thyroid peroxidase (hTPO) antibody was established and its clinical significance was evaluated by measuring levels of serum hTPO antibody. Methods: hTPO was labelled by 125I and the best conditions were selected. The levels of serum hTPO antibody were measured in 37 normal controls and 92 patients with autoimmune thyroid diseases (AITD). Results: Analysis of variables showed that the level of serum hTPO antibody was significant higher in patients with AITD than that in normal controls (F=4.612, P<0.05). Positive rates of serum hTPO antibody in Graves' disease, Hashimoto's thyroiditis and primary myxedema were 66.7%, 95.2% and 72.7%, respectively. Conclusion: This method is of high specificity and sensitivity, good reproducibility, low in nonspecific binding and useful to study and diagnose AITD
