Effect of whole body X-irradiation on the expression of p53, p21, GADD45 and MDM2 proteins in mouse thymocytes

Description
Objective: To investigate the effect of ionizing radiation on the expression of p53, p21, GADD45 and MDM2 proteins of thymocytes in mice. Methods: Flow cytometry (FCM) was used to analyze protein expression of thymocytes for their immunofluorescence stained with monoclonal antibodies. Results: It was demonstrated that after 2 Gy whole body irradiation with X-rays, p53 protein expressions of mouse thymocytes were significantly increased at 1 h to 8 h; p21 at 4 h to 48 h; and MDM2 at 4 h and 8 h. However, no significant change was found in GADD45 protein expression following irradiation. Conclusion: The protein expression of p53, p21 and GADD45 of thymocytes could be induced by ionizing radiation, which might be involved in the molecular pathway of G1 arrest after exposure
