Effects of diphosphonate on kidney calcium content and duodenal absorption of 45calcium

In rats the relationships between EHDP-induced changes in serum calcium concentration, kidney calcium content and duodenal transport of 45calcium were studied. Body weights and kidney weights were similar in all groups. EHDP administration was associated with an increase in serum calcium concentration and kidney calcium content, and a decrease in duodenal 45calcium transport. In the EHDP-treated rats, there was a significant negative correlation between kidney calcium concentration and duodenal 45calcium transport but no correlation between either kidney calcium content and serum calcium concentration (r = 0.116) or between serum calcium concentration and duodenal 45calcium transport (r = 0.02). Further experiments will be needed to determine whether the demonstrated increase in kidney calcium content induced by EHDP administration was the cause of, or was secondary to, inhibition of 1, 25(OH)2D3 synthesis. Die Beziehungen zwischen EHPD-induzierten Veraenderungen der Serumcalciumkonzentration, dem Calciumgehalt der Nieren und dem Transport von 45Ca im Duodenum wurden an Ratten untersucht. Nach Verabreichung von EHPD erhoehte sich die Ca-Konzentration im Serum und in den Nieren, waehrend der 45Ca-Transport in Duodenum zurueckging. Bei den mit EHPD behandelten Ratten bestand eine signifikante negative Beziehung zwischen der Ca-Konzentration in der Niere und dem 45Ca-Transport im Duodenum, jedoch kein Zusammenhang zwischen dem Ca-Gehalt der Niere und der Serum-Ca-Konzentration (r = 0,116) oder zwischen der Serum-Ca-Konzentration und dem 45Ca-Transport im Duodenum (r = 0,02). Es bleibt noch zu klaeren, ob die beobachtete Zunahme des Ca-Gehalts der Niere nach EHDP-Gaben die Ursache fuer die Hemmung der 1, 25(OH)2D3-Synthese oder eine sekundaere Erscheinung war.
