Gamma-irradiation and Poly (ADP-ribosylation) in wound healing

Description
The present work was performed to investigate the role of poly(ADPribosyl)ation (PARylation) in regulating the molecular events of normal wound healing. Moreover, the study was extended to elucidate the effect of PARP-1 inhibition on wound healing accompanied by pre-exposure to gamma-radiation. In order to examine whether PARylation was involved in wound healing; a mouse model of excision wounding with two wounds was induced on the back of each mice. One of the wounds on each mouse was treated with a hydrophilic cream whereas the other wound was covered with hydrophilic cream containing the PARP inhibitor 3-aminobenzamide (3-AB, 50 µM). Wound closure was followed with time and found that 3-AB significantly accelerated the closure of wounds. Similarly to 3-AB, the application of PJ-34 has also stimulated the wound healing process. Furthermore, wounds on PARP-1 Knock Out animals healed faster than wounds on wild type mice, confirming that activation of PARP-1 leads to delayed wound healing
