The protective effect of new haemostatic '8401' on the platelet in irradiated mice and the proliferation of endothelial cells by irradiation

Description
The damange of capillary wall is the important cause of hemorrhange induced by radiation. Experiment showed previously that '8401' : 3- (3, 4, 5-trimet-hoxphenyl) - Propenoic acid ethylene diamine salt increased platelet number of the irradiated mice. The '8401' effect of different dose on influence of prollferation of the endothelial cells was observed. The cultured cells of R22-25 were irradiated with 60Co γ-ray (15GY) at room temperature, then the Cells with an initial density of 3.5 x 105 cells/ml were plated on twelve dishes and 1-5 μg of '8401' were added into two dishes each (the other two dishes were used as control). By the seventh days after platting cells, each dish were trypsinized and counted with a blood counter. The result obtained was as follows: and the cells count of 1 μg, 2 μg and 3μg groupsis 1.86x 105, 2.17 x 105 and 2.21 x 105 cells/dish respectively, but that of control group is 1.23 x 105 cells/dish. the cells count in '8401' goup was much higher than that of control group (p<0.01). The results show that '8401' of a certain dose can stimulate growth of the irradiated endothelial cells and '8401' seems to promote proliferation of the endothelial cells in case of the endothelial cell injury
