RAPD analysis of soybean mutants by space mutation

Description
This report is to research the differences of space-induced soybean mutant lines in DNA level by random amplified polymorphic amplified fragments (RAPD) technology for the sake of providing theoretical basis for crop space-flight mutation breeding. Polymorphism analysis was performed between twelve mutant lines and their original control varieties by random amplified polymorphic amplified fragments (RAPD) technology for the first time. Twelve mutant-lines and their original control varieties were analyzed by randomly selected 70 of RAPD primers. Thirty-nine were polymorphic in 247 DNA bands, which were generated from twelve samples. The total degree of gene polymorphism was 15.8%, and polymorphism rate among samples was between 1.43% and 10%, and fragment amplified was between 250-2100 bp. Therefore, space mutagenesis could result in DNA variation of soybean, and RAPD is also an efficient molecular biological method for space mutagenesis analysis. (authors)
