Input of I-125 radionuclide into antibodies molecule to delta - antigen

Description

In this report there is a detailed modification of the way of I-125 inputting into anti-delta molecule. This modification differs from the standard one by the fact that antibodies to delta-antigen were input into a reaction vial last reducing the time of destructive influence of radionuclide gamma radiation on antibody molecules. We have analyzed possible mistakes occurring in the course of standard method usage. During modified method development of main importance was absolute quantity of chloramine T, inputted in reaction per unit of nuclide radioactivity. Comparison of two ways was conducted by test systems sensitivity obtained for determination of anti-delta where the only variable was chloramine T concentration in reactivity mixture used in tracer manufacture according to calculation for 1 mCi NaI125 at comparing test systems in which there were used tracers made by standard and designed ways. Sensitivity of tracer 2 analysis was higher than analogue factor with tracer-1 1.5-5 times depending on chloramine T concentration used. Advantage data were discussed in relation to the possibility of creating a more sensitive test system for detecting delta-antigen in the blood in patients
