The preparation, animal studies and preliminary clinical evaluation of 99mTc-ECD

Description

The 99mTc-ECD (ethyl cysteinate dimer) is a new brain perfusion imaging agent for SPECT. ECD has been synthesized recently. The 99mTcECD was prepared by ligand exchange reaction with 99mTc-GH (sodium glucoheptonate). The labelling yield was over 94%. The high brain uptake and retention of 99mTc-ECD was observed in monkey, and the tracer clearance was very rapid in blood. But biodistribution studies didn't display high brain uptake in rats with only 0.27% I.D/gram. at 5 min. postinjection. Initial human clinical studies were performed in 30 volunteers. The maximum 99mTc brain activity reached within 1 min. and remained near that level for next 10 min. The brain SPECT images were performed 20 min. after i.v. administration. High quality SPECT images of brain were obtained in all subjects. Preliminary results suggest that 99mTc-ECD is useful for clinical evaluation of rCBF
