Changes of nuclear factor-κB(NF-κB) in cultured bone marrow stromal cells after 60Co γ-irradiation

Objective: To explore the changes in bone marrow hematopoietic microenvironment after exposure to γ-radiation. Methods: Immunohistochemistry, Western blot and electrophoretic mobility shift assay (EMSA)was used. Results: The nuclear translocation was found in cultured bone marrow stromal cells after 60Co γ-irradiation with immunohistochemistry. The expression of NF-κB in the bone marrow stromal cells (BMSCs) on the level of protein was elevated after exposure to 60Co γ-rays at the dose of 8.0Gy with the uses of immunohistochemistry and Western blot. The activities of NF-κB in the BMSCs increased significantly after exposure to γ-radiation by using EMSA. The activity peak was at 4 hours after irradiation. Conclusion: The results suggest that the activation of NF-κB in the BMSCs by irradiation might be involved in the protection of BMSCs and in the recovery of hematopoiesis after γ-irradiation
