In vitro and in vivo measurements of the dissolution parameters of uranium and plutonium mixed oxides in biological environment

During the mixed-oxide fuel fabrication process, inhalation is potentially the main route of internal contamination. The International Commission on Radiological Protection recommends experimental measurement of parameters such as size and dissolution rate for specific industrial compounds. First, we validated the use of PERALS (Photon Electron Rejecting Alpha Liquid Scintillation) for alpha measurement in biological samples which, in some cases, could improve detection limit. We characterised physical chemical properties in terms of size, specific area and activity of 3 different powders: MOX made according to either the MIMAS process, which showed heterogeneous chemical composition, or the SOLGEL, which showed homogeneous chemical composition and industrial PuO2. Their dissolution parameters, fr and ss, as defined in the simplest model proposed by ICRP 66 were measured in vivo, after inhalation in the rat, and in vitro. The statistical variation of these values were expressed as standard deviation. Moreover, in vitro studies demonstrated variation of the ss value depending on the duration of the incubation. We also developed methods to characterise interactions between UO2 particles and phosphate ions which could be involved in the actinide toxicity. (author)

Durant la fabrication des combustibles MOX, (U, Pu)O2, l'inhalation constitue la voie potentielle principale de contamination interne. La Commission Interne, de Protection Radiologique recommande la mesure expérimentale de la granulométrie et des paramètrie de dissolution des différents composés spécifiques d'origine industrielle. Tout d'abord, nous avons validé l'utilisation du PERALS, technique spectrométrique de scintillation liquide alpha, pour l'étude d'échantillons biologiques. Dans certaines conditions, cette méthode permet une diminution significative de la limite de détection des rayonnements alpha après séparation des actinides selon leur état de valence. Ensuite, nous avons étudié des poudres MOX de type MIMAS, de composition chimique hétérogène, de type SOLGEL, de compoeition chimique homogène, et une poudre PuO2. Nous avons caractérisé les propriétés physio-chimiques de ces poudres en terme de granulométrie, de surface spécifique et d'activité, puis, nous avons mesuré leurs paramètres de dissolution in vivo, après inhalation chez le rat, et in vitro. Ces études ont permis de fournir les paramètres fr et ss tels qu'ils sont définis dans le modèle de dissolution le plus simple proposé par la CIPR 66, avec leur variation statistique exprimée en écart type. Par ailleurs, les études in vitro montrent que la valeur de ss peut varier selon le temps d'incubation. Enfin, nous avons développé de nouvelles méthodologies pour mieux appréhender les interactions des ions phosphate avec l'oxyde d'uranium qui interviendraient dans la toxicité chimique de cet actinide.Files
