Ribonucleic acid breakdown in spleen and thymus cells of rats after whole-body X-irradiation

Immediately and 1 and 2 hours after whole-body X-irradiation (GKB, 0.5-10 Gy) the spleen cells of rats were marked in vitro with thymidine 3H (TdR-3H) or uridine-14C(U-14C). After a 40 min. marking phase the breakdown of DNA and RNA was measured by the use of loss of activity of the corresponding nucleic acid fractions. Orientating studies were carried out on RNA breakdown in thymus cells. Results: 1. Incorporation of TdR-3H into DNA was more rapid after irradiation of the whole body and after smaller doses was inhibited more than was incorporation of U-14C into RNA. 2. If spleen and thymus cells were incubated, in contrast, with hydroxy-urea the process of breakdown of nucleic acids affected the RNA more than the DNA. 3. Irradiation intensified RNA breakdown. 4. Increased RNA breakdown can inhibit the restitution of chromatin by damaging nucleic acid synthesis and in this sense also conditions interphase death of irradiation-damaged cells. (orig.)
Abstract (German)
Unmittelbar sowie 1 und 2 Stunden nach Ganzkoerperroentgenbestrahlung (GKB, 0,5-10 Gy) wurden Milzzellen von Ratten in vitro mit Thymidin-3H(TdR-3H) oder Uridin-14C(U-14C) markiert. Nach Abschluss einer 40 min. dauernden Markierungsphase wurde der Abbau der Desoxyribonukleinsaeure (DNA) und der Ribonukleinsaeure (RNA) mit Hilfe des Aktivitaetsverlustes der entsprechenden Nukleinsaeurefraktionen gemessen. Orientierende Versuche galten dem RNA-Abbau in Thymuszellen. Ergebnisse: 1. Durch GKB wurde der Einbau von TdR-3H in die DNA frueher und nach kleineren Strahlendosen gehemmt als die Inkorporation von U-14C in die RNA. 2. Wurden Milz- und Thymuszellen in Gegenwart von Hydroxyharnstoff inkubiert, kam es zu Abbauvorgaengen an den Nukleinsaeuren, welche die RNA staerker betrafen als die DNA. 3. Durch Roentgenbestrahlung wurde der RNA-Abbau verstaerkt. 4. Vermehrter RNA-Abbau koennte ueber eine Stoerung der Nukleoproteinsynthese die Restitution des Chromatins hemmen und in diesem Sinne den Interphasentod strahlengeschaedigter Zellen mitbedingen.
