Effect of Czechoslovak-made aprotinine, Antilysin Spofa, on post-irradiation heamocoagulation disorder

Description

The action was tested of Antilysin Spofa on the coagulation mechanism in rats subjected to whole-body irradiation (206.4 mC/kg). Antilysin Spofa was administered in doses of 75,000 TIU per kg body weight on the 12th day after irradiation. Thirty minutes after administration protracted values of the thromboelastogram were recorded. Subsequently, the effect of repeated administration of Antilysin Spofa to dogs subjected to whole-body irradiation (77.4 mC/kg) was investigated. Antilysin was administered every six hours from the 7th to the 21st days after irradiation, 10,000 TIU per kg body weight. In dogs on the thromboelastograms a protraction of values r and k and diminution of values ma was found. The administration of Antilysin Spofa, however, did not significantly influence the number of white blood cells, platelets, the differential white cell count and the packed cell volume. No changes were recorded in the clotting time in glass and silicon-treated glass. Only in the activated partial thromboplastin time, phasic changes were found after the first administration of Antilysin Spofa and a statistically significant reduction in the coagulation time on the 9th and 11th day after irradiation was observed. From the experiments the conclusion can be drawn that Antilysin Spofa did not favourably influence the coagulation mechanism during the postirradiation disorder. (author)
