Description
The retention of spherical and of irregularly shaped (U, Pu) mixed oxides in rat lung was analyzed after inhalation and intratracheal instillation. Their biological behaviour was relatively independent of particle shape and application route with only a few percent of radioactivity being transferred to other organs. In vivo and in vitro uptake and intracellular distribution in rat and bovine alveolar macrophages were analyzed as dependent on various parameters. In addition, detailed electron microscopic studies were performed demonstrating particles within membrane limited vacuoles as well as lying free in the cytoplasm. Under in vitro conditions the uptake process was finished after a few hours. After differential centrifugation of lung or macrophage homogenates the particles sedimented in the first (1000 g) fraction. (orig.)

Der Einfluss der Partikelform auf das biologische Verhalten von (U, Pu)-Mischoxidpartikeln nach Inhalation oder nach intratrachealer Instillation wurde vergleichend untersucht. Das biologische Verhalten der Teilchen in der Rattenlunge war weitgehend unabhaengig von der Teilchenform und der Applikationsart. Die Translokation und Radioaktivitaet in andere Organe war minimal. Die Aufnahme und intrazellulaere Verteilung der Partikel in Rattenmakrophagen in vivo sowie in Rinderalveolarmakrophagen in vitro wurde in Abhaengigkeit von verschiedenen Parametern untersucht und elektronenmikroskopisch charakterisiert; die Partikel wurden teils frei im Cytoplasma, teils in Phago(-lyso-)somen nachgewiesen. Unter in vitro-Bedingungen war die Partikelaufnahme durch Rinderalveolarmakrophagen innerhalb weniger Stunden abgeschlossen. Nach differentieller Zentrifugation von Lungen- und Makrophagenhomogenaten sedimentierten die Partikel ueberwiegend in der ersten (1000 g) Fraktion. (orig.)
