Labeling of 3H11 With 123I and Its Biodistribution

3H11 was labeled with 123I by Iodogen method,and the labeling product were purified with PD-10 column. The labeling yield and the radiochemical purity of the product was determined by paper chromatography. The biodistribution of 123I-3H11 in normal mice was car ride out as well. The optimal experimental conditions of 123I-3H11 was as follow: Iodogen 10 μg, 3H11 30 μg, Na 123I solution 20 μL (13.3 MBq), PBS 100 μL (pH 7.4, 0.2 mol/L), the normal temperature for 8 min. The labeling yield of 123I-3H11 was 70%-80%. After stored at 4 degree C for 48 h in human serum,the radiochemical purity was more than 92%. The results of biodistribution showed that the clearance of radiolabeled antibody in blood (half time, T1/2) was 12.25±0.25 h, and the radioactivity in the stomach was up taken obviously. The above results indicated that 123I-3H11 appears to show some potential as gastric cancer imaging diagnostic agent. (authors)
