Synthesis of coenzyme A and nicotineamide-adenine dinucleotide labelled with tritium

Description

Isotopic exchange in solution with tritium water and with gaseous tritium and solid-phase reaction of isotopic exchange of NAD with tritium were investigated. For synthesis of labelled with tritium coenzyme A solid-phase reaction of isotopic exchange with gaseous tritium was used. It was determined that 98% of tritium was contained in nicotineamide part of molecule of NAD. In the case of coenzyme A studying of intramolecular distribution of tritium demonstrated that 90% of tritium were localized in adenine fragment

Исследован изотопный обмен в растворе с тритиевой водой и газообразным тритием, а также твердофазная реакция изотопного обмена никотинамидадениндинуклеотида (НАД) с тритием. Для синтеза меченного тритием коэнзима А применяли твердофазную реакцию изотопного обмена с газообразным тритием. В препаратах НАД около 98% трития содержится в никотинамидной части молекулы. В случае с коэнзимом А около 90% трития локализовано в адениновом фрагменте
