Development of a coupled method for demonstrating sister chromatid exchanges and chromosomal marking: application to the detection of radiation-induced chromosomal damage in human lymphocytes

Description

A method is developed, based on the incorporation of 5-Bromodeoxyuridine into the chromosomes of human lymphocytes in culture, making it possible to highlight sister chromatid exchanges and to obtain R band marking (reverse) on mitoses which have already incorporated this substance during two consecutive cycles. The method involves controlled thermal denaturation, followed by coloring with acridine orange. An application to the study of radiation-induced chromosomal damage in human lymphocytes irradiated in G0 revealed an increase and a non-random distribution of sister chromatid exchanges

Abstract (French)

Une methode basee sur l'incorporation de 5-Bromodeoxyuridine dans les chromosomes de lymphocytes humains en culture est developpee. Elle permet de mettre en evidence les echanges de chromatides soeurs et d'obtenir un marquage en bandes R (reverse) sur des mitoses ayant deja incorpore cette substance pendant deux cycles consecutifs. La methode fait intervenir une denaturation thermique menagee, suivie d'une coloration par l'acridine orange. Une application a l'etude des lesions chromosomiques radio-induites dans des lymphocytes humains irradies en G0 a revele une augmentation et une distribution non aleatoire des echanges de chromatides soeurs.
