Microspectrophotometric quantitation of nucleic acid and protein in irradiated epidermis

Description
Nucleic acid and proteins of newborn rat tail epidermis subjected to local X-irradiation were microspectrophotometrically studied. Feulgen, gallocyanine chrom-alum and naphthol yellow S methods were performed for demonstration of DNA, total nucleic acid and proteins respectively. The amount of proteins and total nucleic acid increases concomitantly with reactional acanthosis. However, the proteins and nucleic acid decrease as from day 3 post-irradiation. A tentative interpretation of the results would point to a giantization of the epidermic cells not only caused by aqueous imbition but also by an actual increase of the cellular protoplasm. (orig.)
Abstract (German)
Nucleinsaeuren und Proteine der Epidermis am lokal roentgenbestrahlten Schwanz neugeborener Ratten wurden mikrospektrophotometrisch untersucht. Die Feulgenreaktion, Chromalaun-Gallocyanin-Faerbung und Naphtholgelb-S-Methode dienten zum Nachweis von DNA, Gesamtnucleinsaeuren und Protein. Der Gehalt an Proteinen und Nucleinsaeuren nimmt in gleicher Weise zu wie die reaktive Akanthose, beginnt aber am dritten Tag nach Bestrahlung wieder abzunehmen. Der Versuch einer Deutung der Ergebnisse lenkt die Aufmerksamkeit auf die enorme Vergroesserung der Epidermiszellen, die nicht nur durch Quellung verursacht ist, sondern auch auf wirklicher Zunahme des Zellprotoplasmas beruht.
