Conditioning of mealybug (Hemiptera: Pseudococcidae) by Scanning Electron Microscopy

Description
The cleaning and correct observation of the mealybug specimens was determined by the conditioning methodology. The research was done in the Laboratorio del Centro de Investigacion en Estructuras Microscopicas (CIEMIC) of the Universidad de Costa Rica during the year 2012. A gradual improvement for the observation of the ultrastructures through the Scanning Electron Microscope was evidenced by the implementation of four types of methodologies. Each process was described in detail. The incorporation of 10% xylene (in some cases have been viable using ethanol at 95-100% ) was allowed to remove the wax from the body of the insect, to avoid this the collapse and to observe specific ultrastructures of the individual, they were the best results. The methodology used has reduced the time and costs in future taxonomic research of mealybug. (author)

La limpieza y la correcta observacion de los especimenes de cochinilla harinosa fueron determinadas por la metodologia de acondicionamiento. La investigacion fue realizada en el Laboratorio del Centro de Investigacion en Estructuras Microscopicas (CIEMIC) de la Universidad de Costa Rica durante el ano 2012. Una mejora gradual para la observacion de las ultraestructuras a traves del microscopio electronico de barrido fue evidenciada mediante la implementacion de cuatro tipos de metodologias. Cada proceso fue descrito detalladamente. La incorporacion del 10% de xileno (en algunos casos ha sido viable usando etanol al 95-100%) permitio eliminar la cera del cuerpo del insecto, para evitar el colapso y observar ultraestructuras especificas del individuo, fueron los mejores resultados. La metodologia utilizada ha reducido el tiempo y los costos en futuras investigaciones taxonomicas de la cochinilla harinosa. (autor)Additional details
