Physiology and physiopathology of central type Benzodiazepine receptors: Study in the monkey and in human brain using positron emission tomography

Description

A new non-invasive technique that allows to study in a living subject central type benzodiazepine receptors is developed. A combined approach is applied using a specific positron-emitting radiotracer for the in vivo labelling of the receptors and positron emission tomography allowing, by external detection, a quantitative determination of tissue radioactivity. The radioligand used for the in vivo labelling of benzodiazepine receptors is the antagonist RO 15-1788 labelled with carbon 11. The various stages of the study are described: in vivo characterization in the monkey of central type benzodiazepine receptors; characterization of central type benzodiazepine receptors in human brain using selective molecules for the BZ1 benzodiazepine subclass; demonstration of the heterogeneity of central type benzodiazepine receptors in the brain; study of pathological alteration of benzodiazepine receptors in experimental epilepsy

Abstract (French)

On a developpe, une methode nouvelle permettant d'etudier, dans un organisme vivant et de facon atraumatique, les recepteurs des benzodiazepines de type central. Cette methode combine l'utilisation d'un radiotraceur specifique, emetteur de positons, pour le marquage in vivo des recepteurs et celle de la tomographie par emission de positons afin de pouvoir effectuer, par detection externe, la quantification des rayonnements emis. La mise au point de cette methode a ete effectuee en plusieurs etapes: - realisation d'une methode de marquage, par un radioisotope emetteur de positons (carbone 11), d'un ligand hautement specifique des recepteurs des benzodiazepines, le RO 15-1788, antagoniste specifique de type central, - caracterisation in vivo chez le singe, des recepteurs des benzodiazepines de type central, - caracterisation de ces memes recepteurs dans le cerveau humain, - en utilisant des molecules selectives de la sous-classe BZ1 des recepteurs des benzodiazepines, demonstration de l'existence, au niveau cerebral, d'une certaine heterogeneite des recepteurs des benzodiazepines, de type central. Cette methode a permis d'etudier la physiopathologie des recepteurs des benzodiazepines dans differents modeles experimentaux d'epilepsie
