Determination of the mechanism of early endocytosis of Clostridium perfringens phospholipase C in two fibroblast cell lines
Description
The mechanism of early endocytosis of Clostridium perfringens phospholipase C toxin (Cp-PLC) was determined in Don Q cell lines (Chinese hamster lung fibroblasts) and HSF (primary explant of human skin fibroblasts). The mechanism of entry of the Cp-PLC toxin into Don Q fibroblasts through of mediated endocytosis caveolin and/or clathrin was determined by immunofluorescence. The mechanism of entry of the Cp-PLC toxin into HSF fibroblasts through of caveolin-mediated endocytosis was determined by immunofluorescence. The binding of the Cp-PLC toxin to the membrane of Don Q fibroblasts even at 4 degrees celsius was determined by immunofluorescence. The preparations were observed under a fluorescence microscope and then under a confocal microscope to confirm the placement of the toxin with caveolin or clathrin. At least 25 photographs were taken by sheet at the point of greatest colocalization

El mecanismo de endocitosis temprana de la toxina fosfolipasa C de Clostridium perfringens (Cp-PLC) fue determinado en las lineas celulares Don Q (fibroblastos de pulmon de hamster chino) y HSF (fibroblastos de explante primario de piel humana). El mecanismo de entrada de la toxina Cp-PLC a los fibroblastos Don Q por medio de endocitosis mediada por caveolina y/o claratina fue determinado mediante inmunofluorescencia. El mecanismo de entrada de la toxina Cp-PLC a los fibroblastos HSF por medio de endocitosis mediada por caveolina fue determinado mediante inmunofluorescencia. La union de la toxina Cp-PLC a la membrana de los fibroblastos Don Q aun a 4 grados centigrados fue determinada mediante inmunofluorescencia. Las preparaciones fueron observadas en un microscopio de fluorescencia y posteriormente en el microscopio confocal para confirmar la colocalizacion de la toxina con caveolina o clatrina.
