The LIGOP method for routine ultrastructural autoradiography. A combination of single grid coating gold latensification and phenidon development
Description
A detailed description of the whole procedure of the LIGOP method (loop-interference-gold latensification-phenidon method) is presented including the following stages: sample processing; coating; photographic processing; staining. This procedure results in dot-like, round shaped silver grains of regular diameter easy to identify and count for autoradiographic quantitation. Their size may be varied according to the temperature and duration of treatment of gold latensification and development. Occasionally, very small or large and irregular grains are obtained. This is usually due to poor coating and the grids should be discarded. Nevertheless, when the most favorable conditions are met and with some practice, the LIGOP method appears as a reliable, consistent and convenient method
