New cyclam-based chelate derivatives for cancer diagnostic and/or therapeutic applications
Description
The work consisted in the development of chelates based on π-extended picolinate cyclam derivatives giving access to luminescent complexes for applications in medical imaging and/or therapy. For a specific use for biphotonic microscopy, an Yb3+ reinforced cyclam complex functionalized by two π-extended picolinate units has been synthesized. The simultaneous presence of a lanthanide ion emitting in the NIR and an antenna allowing two-photon absorption, gave rise to cellular images recorded in the NIR-to- NIR channel. The luminescent properties of π-extended picolinate antennas have also been used for the development of bimodal PET/optical imaging probes. Cyclam-based ligands have been functionalized with a picolinate fluorophore and have been used for the complexation of Cu2+ which has a β+-emitting isotope. If the radiolabeling of the ligands with 64Cu2+ could be carried out, spectroscopic studies of the Cu2+ complexes demonstrated that the metal inhibits the luminescent properties of the fluorophore. However, preliminary studies have shown that Ga3+, which also has a β+-emitting isotope, may be a relevant alternative to Cu2+. Finally, the substitution of the π-conjugated system by the incorporation of a dibromobenzene unit gives access to a photosensitizer capable of generating cytotoxic singlet oxygen for a use in photodynamic therapy (PDT). This type of photosensitizer can also be introduced on the picolinate moiety to give Cu2+ chelates for an application in TEP/PDT theranostic. Again, the influence of the metal center on the photophysical properties of the resulting complex could be highlighted. (author)

Le travail de these a consiste au developpement de chelates derives du cyclam N-fonctionnalises par des groupements picolinate π-etendus donnant acces a des complexes luminescents pour des applications en imagerie medicale et/ou en therapie. Pour une application en microscopie biphotonique, un complexe d'Yb3+ base sur le cyclam renforce substitue par deux unites picolinates π-etendues a ete synthetise. La presence simultanee d'un ion lanthanide emettant dans le NIR et d'une antenne permettant l'absorption a deux photons, a donne lieu a des images cellulaires par microscopie biphotonique dans le canal NIR-vers- NIR. Les proprietes luminescentes des antennes picolinate π- conjuguees ont egalement ete mises a profit pour le developpement de sondes bimodales TEP/imagerie optique. Des ligands a base de cyclam ont ete fonctionnalises par un fluorophore picolinate et ont servi a la complexation du Cu2+ qui possede un isotope emetteur β+. Si le radiomarquage des ligands au 64Cu2+ a pu etre realise, les etudes spectroscopiques des complexes de Cu2+ ont montre que le metal inhibe les proprietes luminescentes du fluorophore. Toutefois, des etudes preliminaires montrent que le Ga3+, possedant egalement un isotope emetteur β+, s'avere etre une alternative pertinente au Cu2+. Enfin, la substitution du systeme π-conjugue par une unite dibromobenzene permet d'avoir acces a un photosensibilisateur capable de generer de l'oxygene singulet cytotoxique pour une application en therapie photodynamique (PDT). Ce type de photosensibilisateur peut lui aussi etre introduit sur l'entite picolinate des derives du cyclam donnant acces a des chelates de Cu2+ pour une application en theranostique TEP/PDT. A nouveau, l'influence du metal sur les proprietes photophysiques des complexes a pu etre mise en avant. (auteur)
