Cutaneous permeability 'in vitro'

Description

Using the isolated skin technique, a comparative study was undertaken in post mortem specimens of human skin and from biopsy specimens of monkeys, pigs, rabbits, guinea-pigs and hairless rats. With 14C-labeled methanol, the authors have experimentally observed that the penetration was quite similar through the skin of men and hairless rats. 14C-methanol was less penetrant in all other species and was minimal in pigs. Mathematical analysis of experimental data confirmed this observation. Microscopic autoradiographic examinations indicated 14C-localizations. This preliminary investigation determined an animal model for estimating skin permeability and will be used for further comparative and systematic experiments with industrial solvents

Abstract (French)

Les auteurs ont utilise la technique de la peau isolee pour l'etude de la permeabilite cutanee chez l'homme (prelevement post-mortem) et chez plusieurs especes animales (primate, porc, lapin, cobaye, rat mutation atrichis). Avec le 14C-methanol comme produit de reference, ils ont experimentalement observe que le rat mutation atrichis avait une permeabilite voisine de celle de l'homme alors que celle du porc etait minimale. Cette constatation a ete confirmee par l'analyse mathematique du phenomene. Des autohistoradiographies ont ete effectuees afin de localiser le radio-carbone. Les resultats sont discutes par rapport aux donnees de la litterature. Cette etude prealable permettra une etude systematique comparative des solvants industriels
