Radiochemical and biological evaluation of 153Sm-EDTMP, complemented with a study of stability

We have evaluated the radiochemistry purity of 153Sm-EDTMP, in four different chromatography systems: water/ITLC-SG; ammonia/ITLC-SG; NH3: MEOH: H2O (0.2:2.0:4.0)/whatman No3 and by chromatography in a packed mini-column with sephadex G25, being eluded with NaCl 0,9%. The four chromatography systems reported a major radiochemical purity of 99%, but in the ammoniacal - alcoholic system the resolution is poor; of the four systems is chosen the mini column to be a rapid and simple technique. For the biological evaluation we made biodistributions in healthy animals in different times: mice to 30 minutes; 1; 2; 3; 4 and 6 hours and in rats to the 3 and 24 hours post-injection; we determine that the ideal biological model is the rat, that the time of sacrifice is 3 hours post-injection and that the minimum percentage of dose injected to femur is 1%, for both. These evaluations were complemented with a study of stability on real time of the 153Sm-EDTMP, storing between 2 to 8oC, having determined the radiochemical purity by mini-column for ten days, obtaining a stability until the 4 days with a major radiochemistry purity of 99%.(author)
