The role of exopolysaccharide-producing-bacteria (Rhizobium sp.) in sunflower rhizospheric-soil aggregation: effect of inoculation on soil structuration and plant nutrition

Description

The bacterial strain Y AS34, selected for exopolysaccharide (EPS) production, was isolated from the rhizoplane of sunflower (Helianthus annuus) in a Dystric Cambisol. From cartography and sequencing of the 16S rDNA gene it was concluded that strain Y AS34 belongs to the genus Rhizobium. The gelatinous EPS produced by this strain was different from succinoglycan usually produced by bacteria belonging to the genus Rhizobium. In the presence of 1 % (w/w) of the EPS purified from strain Y AS34, the water holding capacity of soil from St-Florent-sur-Cher was enhanced by 50%. Inoculation of sunflower seeds and soil with strain Y AS34 led to a significant increase of root-adhering soil mass per root biomass (+80%), under both, water stress and normal water supply conditions. Concomitantly with this bacterial effect on the mass of root-adhering soil, an increase of soil macro-porosity (pore diameter between 5 and 50 mm) was observed. In addition to these physical changes of root-adhering soil, nitrogen uptake by sunflower was significantly increased after inoculation with Rhizobium sp. Y AS34 in the presence of 1 N-nitrates. Strain Y AS34 seems to modify the soil structure around the root system in a way that the water supply of sunflower and its nitrogen uptake is increased counteracting the effect of water deficit on sunflower growth. Abstract (French)

La souche Y AS34, selectionnee pour la qualite et la quantite d 'exopolysaccharide (EPS) qu'elle produit, a ete isolee de la surface de racines de tournesol cultive sur un sol sablo-limono-argileux de St- Florent-sur-Cher (18, France). La cartographie et le sequencage du gene codant pour l'ARNr 16S ainsi que la presence du gene nodD ont permis d'identifier cette souche Y AS34 au genre Rhizobium. Elle se caracterise par la production d'un EPS gelifiant, poly-electrolyte, de structure et de composition differentes des succinoglycanes habituellement rencontres chez les bacteries du genre Rhizobium. L'ajout de cet EPS, a une teneur de 1 %, au sol de St-Florent-sur-Cher s'est traduit par l'augmentation de sa capacite de stockage d'eau (+50%). L'inoculation du sol et des semences de tournesol par la souche Y AS34 a permis d'augmenter significativement la taille de la population de Rhizobium sp. Y AS34 dans la rhizosphere et sur les racines de tournesol (+50% de la microflore productrice d'EPS). Cette colonisation rhizospherique et racinaire s'accompagne d'une augmentation de la biomasse racinaire et de la masse du sol adherent par unite de masse de matiere seche racinaire (SA/PA) (en moyenne +80%) et une modification de la structuration de cette fraction de sol: augmentation de la macro-porosite dont les rayons d'entree des pores sont compris entre 5 et 50 mm. Ces modifications qualitatives et quantitatives de l'environnement physique des racines de tournesol, dues a l'inoculation, ont ete concomitantes d'une augmentation a la fois de la teneur en azote au niveau du sol adherent et des exportations de cet element vers la plante ainsi qu'a une valorisation plus importante de l'engrais azote apporte en debut de culture, par comparaison avec des plantes non inoculees.
