Optimization of a method by liquid chromatography of high resolution to determine residues of ethilenthiourea in samples of tomato
Description
A method was optimized to determine the present residues of ethilenthiourea in samples of tomatoes. The method consisted of three stages: a extraction of ultrasonic bath with methanol; a cleaning of the extract through a glass column of 11 mm of diameter stuffed with 2,5 g of neutral aluminium's mixture and activated coal (97,5:2,5) and 2,5 g of pure-neutral aluminium, it is dissolved with 250 ml of methanol. The third stage was of quantification by HPLC in a C18' column with a methanol and water mixture (90:10) like a mobile phase to an flow of 2,0 ml/min. and with UV detection to 232 nm. The retention's time under theses conditions was of 2,15 minutes. The merit's parameters of the method were determined, proving and lineal sphere between 1,0 and 28,0 (g/ml of ETU; some quantification and detection limits have been calculated by the method of Hubaux and Vos (22) of 0,153 and 0,306 mg/ml respectively and a recuperation of 84%. Se optimizo un metodo para determinar los residuos de Etilentiourea presentes en muestras de tomates. El metodo consistio de tres etapas: una extraccion en bano ultrasonico con metanol; una limpieza del extracto a traves de una columna de vidrio de 11 mm de diametro rellena con 2,5 g de mezcla de alumina neutra y carbon activado (97,5:2,5) y 2,5 g de alumina neutra pura, eluyendo con 250 mL de metanol. La tercera etapa fue la de cuantificacion por HPLC en una columna C18' con una mezcla agua y metanol (90:10) como fase movil a un flujo de 2,0 mL/min. y con deteccion UV a 232 nm. El tiempo de retencion bajo estas condiciones fue de 2,15 minutos. Se determinaron los parametros de merito del metodo, comprobandose un ambito lineal entre 1,0 y 28,0 (g/mL de ETU; unos limites de deteccion y de cuantificacion que se calcularon por el metodo de Hubaux y Vos (22) de 0,153 y 0,306 mg/mL respectivamente y una representacion de 84%.
