Interaction of Cosup(III) complexes with radiation on 'in vitro' mammalian cells and on molecular systems

Description

Cosup(III) complexes could be considered as potential radiosensitizers in dependence of the electronic configuration of the metal. Due to the high redox state that causes its powerful electrophilia they possibly act through electron-affinic mechanisms. In this paper the effect of [Co(NH3)5Cl]Cl2 on TC.SV-40 hamster cells is reported, in aerobic and hypoxic conditions, and its action, at a non-toxic concentration, in a combined treatment with X-rays. Results show the small protection induced by the product in the survival curves. As a contribution to the knowledge of its possible mechanism of action some molecular proofs and the interaction test with -SHNP cellular groups are made. A high degree of reaction is observed as is the case with good radiosensitizers but at molecular level the behaviour of the complex formed between DNA and the product reveals mutual protection due to their corresponding radiodegradation. Abstract (German)

Cosup(III)-Komplexe koennen durch die Elektronenkonfiguration des Metalls als potentielle radiosensitizer betrachtet werden. Durch den hohen Redoxzustand, der eine starke Elektrophilie bewirkt, wirken sie moeglicherweise durch elektronenaffine Mechanismen. Die Arbeit berichtet ueber die Wirkung von Co(NH3)5ClCl2 auf TC.SV-40-Hamsterzellen unter aeroben und hypoxischen Bedingungen und ueber seine Wirkung in nichttoxischer Konzentration in Kombination mit Roentgenstrahlung. Die Ergebnisse zeigen die schwache Schutzwirkung des Produkts in den Ueberlebenskurven. Zur besseren Kenntnis des Wirkungsmechanismus werden molekulare Nachweise und ein Test der Wechselwirkung mit -SHNP-Zellgruppen durchgefuehrt. Wie bei guten Radiosensitizern wird eine starke Reaktion beobachtet; auf der molekularen Ebene zeigt das Verhalten des DNS/Produktkomplexes eine gegenseitige Schutzwirkung aufgrund der entsprechenden Strahlungszersetzung.
