Description
The flow cytometry techniques allow the analysis and sorting of living biologic cells at rates above five to ten thousand events per second. After a short review, we present in this report the design and development of a 'high-tech' apparatus intended for research laboratories and the experimental results. The first part deals with the physical principles allowing morphologic and functional analysis of cells or cellular components. The measured parameters are as follows: electrical resistance pulse sizing, light scattering and fluorescence. Hydrodynamic centering is used, and in the same way, the division of a water-stream into droplets leading to electrostatic sorting of particles. The second part deals with the apparatus designed by the 'Commissariat a l'Energie Atomique' (C.E.A.) and industrialised by 'ODAM' (ATC 3000). The last part of this thesis work is the performance evaluations of this cyto-meter. The difference between the two size measurement methods are analyzed: electrical resistance pulse sizing versus small-angle light scattering. By an original optics design, high sensitivity has been reached in the fluorescence measurement: the equivalent noise corresponds to six hundred fluorescein isothiocyanate (FITC) molecules. The sorting performances have also been analyzed and the cell viability proven. (author)

La cytometrie en flux est une technique qui permet l'analyse et le tri de cellules biologiques vivantes a des cadences pouvant atteindre cinq a dix mille evenements par seconde. Apres un rapide historique, nous presentons dans ce memoire la conception et le developpement d'un appareil 'haut-de-gamme' ainsi que les resultats experimentaux. La premiere partie expose les principes physiques permettant l'etude morphologique et fonctionnelle des cellules ou des constituants cellulaires. Les parametres accessibles sont: la mesure electrique du volume, la diffusion de la lumiere et la fluorescence. Le principe du centrage hydrodynamique y est egalement presente, ainsi que le fractionnement d'un jet d'eau en gouttelettes permettant le tri electrostatique des particules. Dans la deuxieme partie, nous decrivons l'appareil que nous avons concu au Commissariat a l'Energie Atomique (C.E.A.) et industrialise par la Societe ODAM (ATC 3000). La derniere partie de cette these est consacree a l'evaluation des performances de ce cytometre. Nous y analysons la difference entre les deux methodes de mesure de la taille: volume par la methode electrique compare a la diffusion aux petits angles. Nous montrons que notre optique originale a une sensibilite elevee pour la mesure de fluorescence de sondes biologiques; le bruit obtenu est equivalent a six cents molecules d'isothiocyanate de fluoresceine (FITC). Nous analysons egalement les performances du tri et nous prouvons qu'il respecte la viabilite cellulaire. (auteur)Files
