Synthesis and Comparison of [y32p]-atp Labeled Nucleotide between Domestic and imported product

Description
Molecular biology research is growing rapidly today, and it's a good opportunity to use radioisotopes to explore a variety of processes such as physiology, pathology and imaging techniques. The most widely used radioisotope in molecular biology research is 32p in the form of short-chain nucleotides e.g. nucleotide labeled with 32p Adenosine Triphosphate ([y-32P]-ATP). The nucleotide labeled [y-32p]-ATP is synthesized through an enzymatic reactions using DL-glyceraldehyde phosphate, adenosine diphosphate (ADP) and 32P, with glyceraldehyde 3-phosphate dehydrogenase, 3-phosphoglycerate kinase and lactate dehydrogenase enzymes. Separation of the synthesis results was carried out using of DEAE-Sephadex chromatography column. The labeled nucleotides were characterized using thin layer chromatography (TLC) plates. And gave radiochemical purity test results of 99.49 % at Rf 0.244. The product of synthesis of la bled nucleotide [y-32P]-ATP was then compared with imported product from Hartmann Analytic German. The result showed that the local product has radiochemical purity 99,49 % at RF 0.244 while imported product give radiochemical purity 97,24 % at RF 0.204. It was concluded that the radiochemical purity of local product was better compared to imported product. (author)
