Investigations of the radiosensitivity of erythropoietin

Description

The radiosensitivity of erythropoietin (ESF) in the plasma of anaemic rats was determined by irradiating the plasma containing erythropoietin with X-rays or γ-rays of 0.1 Mrd to 8.4 Mrd. Inactivation of the erythropoietin was measured by means of the radioiron test via the stimulation of erythropoiesis in polyglobulic mice. At doses of 0.1 Mrd and 1.1 Mrd, no inactivation of ESF was observed, while after irradiation with 8.4 Mrd ESF inactivation amounted to 30% of the initial value. The dose-effect curve results in a radiation dose D37 of 6.9 Mrd. From this value, the molecular weight of erythropoietin in the plasma of anaemic rats was determined to be 84,000 on the basis of hit theory. (orig./MG)

Abstract (German)

Es wurde die Strahlenempfindlichkeit des Erythropoetins (ESF) im Plasma anaemischer Ratten bestimmt. Das erythropoetinhaltige Plasma wurde mit Roentgen- bzw. Gammastrahlen im Dosisbereich von 0,1 Mrd bis 8,4 Mrd bestrahlt. Die Inaktivierung des Erythropoetins wurde ueber die Stimulierung der Erythropoese bei polyglobulen Maeusen mit Hilfe des Radioeisen-Tests gemessen. Waehrend bei den Strahlendosen von 0,1 Mrd und 1,1 Mrd keine Inaktivierung des ESF beobachtet werden konnte, betrug die Inaktivierung des ESF nach Bestrahlung mit 8,4 Mrd 30% des Ausgangswertes. Aus der Dosis-Effekt-Kurve ergibt sich eine Strahlendosis D37 zu 6,9 Mrd. Aufgrund von treffertheoretischen Ueberlegungen wurde daraus das Molekulargewicht des Erythropoetins im Plasma anaemischer Ratten zu 84.000 berechnet. (orig./MG)
