Gamma-ray-induced DNA single strand break and their repair in V79 cells and thymocytes
Description
The gamma-ray-induded single strand breaks of DNA molecules and their rejoining in cultured Chinese hamster lung fibroblasts (V79 cells) and thymocytes isolated freshly from mouse thymuses were detected by KOhn's alkaline elution method. The percentage of DNA remaining on the filter membrane was inversely proportional to the radiation dose within the ranges of 1-30 Gy and for V79 cells, and of 1-10 Gy for mouse thymocytes. V79 cells were capable of rejoining the breaks and the rate of rejoining was dependent on the radiation dose. The repair of damaged DNA could carry out in V79 cells cultured in TC199 medium with or without serum, but it could not be observed in mouse thymocytes cultured in RPMI 1604 without serum or TC199 medium without serum
