Radioimmunological determination of total thyroxine in the serum

Description

A radioimmunological method to determine total thyroxine in a serum sample is described. The method is as follows: trichloracetic acid and sodium hydroxide are mixed with the sample; radioactive thyroxine is added to the mixture, which is left to reach equilibrium then placed in contact with a resin sponge consisting of a polyurethane foam with intercommunicating cells containing a strongly basic anion exchange resin; the mixture and the resin sponge are incubated, the initial radioactivity of the mixture and resin sponge combination is measured with an appropriate detection system, then the resin sponge is removed from the mixture, washed and its residual radioactivity measured

Abstract (French)

On presente une methode de dosage radio-immunologique de la thyroxine totale dans un echantillon de serum. Elle consiste a melanger de l'acide trichloracetique et de l'hydroxyde de sodium a l'echantillon de serum pour former un melange; a ajouter de la thyroxine a marquage radio-actif au melange; a laisser le melange s'equilibrer; a placer au contact du melange une eponge de resine constituee d'une mousse de polyurethane a cellules intercommunicantes renfermant une resine echangeuse d'anions fortement basique; a incuber le melange et l'eponge de resine; a mesurer la radio-activite initiale de la combinaison du melange et de l'eponge de resine avec un dispositif de detection approprie; a retirer l'eponge de resine du melange; a laver l'eponge de resine; et a mesurer la radio-activite residuelle de l'eponge de resine
