UV dosimetry in pollen of Pinus silvestris and stimulation studies of pollen tube growth after irradiation with UV and ionizing radiations

Pollen tube growth after exposure to UV- and ionizing radiation was investigated in Pinus silvestris as a function of different parameters. The preconditions for this are an exact UV dosimetry and the conversion of the UV dose of [erg] into [rad]. In spite of the fact that the calculation methods are theoretically already well known, the experimental determination of the absorbed energy dose, in particular the absorption factors, was complicated by the morphological structure of the pollen. The total-irradiation experiments showed that the stimulation and inhibition of pollen tube growth is highly dependent on the dose/dose rate region chosen. Stimulation is highly dependent on the dose rate after UV irradiation as well as after exposure to ionizing radiation. When the dose rates are very small, the values are practically the same as in non-irradiated controls; with increasing doses rate, stimulation increases, whereas at very high dose rates only growth inhibition is observed. After exposure to ionizing radiation, the quantum energy or wavelength of the radiation has no effect on tube growth; after UV irradiation, on the other hand, there is hardly any stimulation at short wavelengths. As these observations were not sufficient for drawing conclusions on the site of the stimulatory action, partial UV exposure with separate irradiation of the two nuclei and the cytoplasm was also carried out. Stimulation was found in each of the three irradiation sites; although nuclei and cytoplasma react differently and also differ in the initial reaction stages, there is always an anhancement of pollen tube growth. These findings are compared with some of the current hypotheses for an explanation of the stimulatory effect. (orig.)

Das Schlauchwachstum von Pollen von Pinus silvestris wurde nach UV- und ionisierender Bestrahlung in Abhaengigkeit verschiedener Parameter der Strahlung untersucht. Voraussetzung zu einem solchen Vergleich war eine exakte UV-Dosimetrie und die Umrechnung der UV-Dosis von [erg] in [rad]. Trotz der theoretisch schon lange bekannten Berechnungsmethoden war die experimentelle Bestimmung der absorbierten Energiedosis, insbesondere der Absorptionsfaktoren, durch den morphologischen Aufbau der Pollen sehr erschwert. Bei den Totalbestrahlungsexperimenten wurde nachgewiesen, dass die Stimulation und Hemmung des Pollenschlauchwachstums stark von den gewaehlten Dosis/Dosisleistungsbereichen abhaengt. Die Stimulation zeigt eine ausgepraegte Abhaengigkeit von der Dosisleistung sowohl nach UV- als auch nach ionisierender Bestrahlung. Bei sehr kleinen Dosisleistungen findet man praktisch keine Abweichung von den unbestrahlten Kontrollen, mit zunehmender Dosisleistung nimmt die Stimulation ebenfalls zu und bei sehr hohen Dosisleistungen findet man fast nur noch Wachstumshemmungen. Die Quantenenergie bzw. Wellenlaenge der Strahlung beeinflusst nach ionisierender Bestrahlung das Schlauchwachstum nicht unterschiedlich, dagegen konnte nach UV-Bestrahlung gezeigt werden, dass bei kuerzeren Wellenlaengen kaum noch Stimulation auftritt. Da hieraus noch keine Schluesse auf den Wirkungsort der Stimulation gezogen werden konnten, wurden UV-Partialbestrahlungen durchgefuehrt, bei denen die beiden Kerne und das Cytoplasma getrennt bestrahlt werden konnten. Dabei ergab sich, dass Stimulation fuer jeden dieser drei Bestrahlungsorte erhalten wird, wobei die Kerne und das Cytoplasma verschieden empfindlich reagieren und sich auch in den anfaenglichen Reaktionsstufen unterscheiden, jedoch stets eine Zunahme des Pollenschlauchwachstums zeigen. Die vorliegenden Resultate werden mit einigen bisher bekannten Hypothesen zur Erklaerung der Stimulation verglichen. (orig.)
