Studies on granulocyte kinetics by means of 32P-labelled diisopropylfluorophosphate and 51Cr

Granulocyte kinetics were determined in vitro and in vivo using 32P-labelled diisopropylfluorophosphate (DF32P) or 51Cr. Various parameters such as total blood granulocyte pool, circulating granulocyte pool, marginal granulocyte pool, daily granulocyte exchange rate, and half-life of granulocytes were assessed in vitro, whereas in the in vivo studies the half-life of granulocytes, the reserve of bone marrow concerning myclocytes, metamyelocytes, juvenile and polymorphonuclear neutrophils could be determined. The combination of both the procedures revealed the half-life, pool sizes, and exchange rates of the proliferating myelocyte compartment in bone marrow and nature blood granulocytes. Using 51Cr in the half-life determination of granulocytes the surface activities of various organs such as heart, liver, spleen, and lungs can chiefly be determined thus indicating the degradation or the storage of cells in certain areas of the body. Besides the normal values those data are mainly reported, which were revealed by the in vitro labelling with DF32P and 51Cr in chronic myeloid leukemia, osteomyelofibrosis, osteomyelosclerosis, and hypersplenism, resp.
