Modification of survival of gamma irradiated mice by adenosine nucleotides

Description

The administration prior to irradiation of adenosine triphosphate (ATP) or other adenosine nucleotides, singly or in combination, increased the radioresistance of mice. Post-irradiation treatment with the adenosine nucleotides had no effect on the survival of the irradiated mice. Dose reduction factors of 2.32 could be obtained by pretreatment of mice with the following combination of protective agents: S-2(4-aminobutylamino)ethyl phosphorothioic aced (WR 2822), cysteamine (MEA) and ATP. Since cyclic AMP levels were unchanged in the spleen or gut by administration of cysteamine and other protectors it is unlikely that the increase in protection was due to changes in cyclic AMP levels. The calcium salt of ATP provided a higher level of protection than the ATP alone, indicating that the protective mechanism of ATP is probably not related to anoxia. (orig.)

Abstract (German)

Adenosintriphosphat (ATP) und andere Adenosinnucleotide, einzeln oder im Gemisch, erhoehten die Radioresistenz von Maeusen, sofern die Verabreichung vor dem Bestrahlen erfolgte. Adenosinnucleotidbehandlung nach dem Bestrahlen hatte jedoch keinen Einfluss auf die Ueberlebensquote von Maeusen. Dosisreduktionsfaktoren von 2.32 wurden erhalten, wenn die Maeuse mit der folgenden Kombination von Schutzstoffen vorbehandelt wurden: S-2(4-Aminobutylamino)aethyl-Phosphorthiosaeure (WR 2822), Cysteamin (MEA) und ATP. Die erhoehte Schutzwirkung kann kaum auf Unterschiede in der Konzentration von zyklischer AMP zurueckgefuehrt werden, da die Konzentration von zyklischer AMP in Milz und Darmkanal bei der Verabreichung von Cysteamin und anderen Schutzstoffen konstant bleibt. Das Kalziumsalz von ATP zeigte eine hoehere Schutzwirkung als freies ATP, ein Hinweis dass der Mechanismus der Schutzwirkung wahrscheinlich nicht im Zusammenhang mit Sauerstoffmangel (Anoxia) steht.
