Probing protein structure with... neutrons
Description
In this thesis, I present a review of my work in neutron scattering applied to protein structural studies, which corresponds to approximately the last six years of my own research. I will first introduce the scientific context of my work by situating SANS and SNR in the landscape of protein structural biology (Chapter 1). As these techniques are relatively little used in biology, I will review the principles of both techniques. In particular, I will discuss their complementarity with higher resolution techniques and the advantages of the contrast variation method. I will then deal more specifically with two themes in which I have been able to use contrast variation to determine, in collaboration with biologists, the structure of proteins in complex environments: (i) biological assemblies composed of proteins interacting with a partner, in particular membrane lipids (Chapter 2); (ii) proteins subjected to highly concentrated environments or under pressure (Chapter 3). The low-resolution protein structures obtained in these studies have enabled us to gain a better understanding of the function of these proteins, with a view to possible applications, notably in the biomedical field. Abstract (French)
Je presente dans ce memoire un bilan de mes travaux en diffusion de neutrons appliquee a des etudes structurales de proteines, qui correspondent environ aux six dernieres annees de ma recherche propre. J'introduirai d'abord le contexte scientifique de mes travaux en situant le SANS et le SNR dans le paysage de la biologie structurale des proteines (Chapitre 1). Etant donne que ces techniques sont relativement peu utilisees en biologie, je rappellerai les principes des deux techniques. En particulier, je parlerai de leur complementarite par rapport aux techniques a plus haute resolution et de l'atout de la methode de la variation de contraste. Je traiterai ensuite plus specifiquement deux thematiques, pour lesquelles j'ai pu mettre en pratique la variation de contraste pour determiner, en collaboration avec des biologistes, la structure de proteines dans des environnements complexes: (i) des assemblages biologiques composes de proteines en interaction avec un partenaire, en particulier des lipides membranaires (Chapitre 2); (ii) des proteines soumises a des environnements tres concentres ou sous pression (Chapitre 3). Les structures proteiques a basse resolution obtenues dans ces etudes ont permis de mieux comprendre la fonction de ces proteines, en vue d'applications possibles, dans le biomedical notamment
