Drug uptake (DAPI) of trypanosomes (T. brucei) and antitrypanosomal activity in vitro, in culture and in vivo studied by microscope fluorometry, chromatogram spectrophotometry and radiotracer techniques

Description

The present study had the following objectives: 1) Investigation of the specific binding and location of the diamidine DAPI within trypanosomes by fluorescence microscopy. 2) Development and standardization of a microscope fluorometry technique for measuring DAPI uptake of single trypanosomes. 3) Determination of the effect of incubation media, exposure time, and drug concentration on DAPI uptake of single trypanosomes. 4) Development of a technique applicable for quantitative fluorescence chemical analysis of DAPI uptake of trypanosomes. 5) Determination of drug uptake of trypanosomes using 14C labelled DAPI. 6) Comparison of the values obtained by the three methods.
