Observations on the effect of radiotherapy, followed by injection of pig lymph node cells, in reducing the number of pulmonary tumors induced by intravenous injection of tumor cells into isogenic mice
Description
Pulmonary tumors were produced in A. strain mice by intrevenous injection of A. strain mammary carcinoma cells. The mesenteric lymph nodes of pigs were immunized by implantation of fragments from the same tumors into the pig mesentery. Tumor-immune pig lymph node cells when injected IV 7 days after tumor cells did not reduce the number of tumors, counted on day 14. However, when preceded by 200 rad thoracic irradiation on day 3 (which increased the number of pig cells settling in the lungs) tumor-immune cells given IV reduced the number of tumors compared with the effect of irradiation alone, or in combination with nonimmune pig cells. When tumor-immune pig cells were injected IP on day 7 (following thoracic irradiation on day 3), no antitumor effect was observed. Thus immediate pig cell/tumor cell contact is important in order to obtain an antitumor effect. Pig cells immunized against a human bladder carcinoma did not reduce pulmonary tumor formation by one of the mouse tumors. (orig.)

Lungentumore wurden bei A-Maeusen durch IV-Injektion von A-Brustkrebszellen erzeugt. Die mesenterischen Lymphknoten von Schweinen wurden durch Implantation von Fragmenten des gleichen Tumors im Schweinemesenterium immunisiert. Durch IV-Injektion von tumorimmunen Schweinelymphknoten Zellen 7 Tage nach Injektion der Tumorzellen wurde die Tumorzahl nicht beeinflusst (Zaehlung am 14. Tag). Bestrahlte man dagegen am 3. Tag mit 200 rad am Thorax (wodurch sich die Zahl der in der Lunge festgesetzten Schweinezellen erhoehte), so bewirkte die IV-Injektion von immunen Schweinezellen einen Rueckgang der Tumorzahl im Vergleich zur Bestrahlung allein oder in Verbindung mit nichtimmunen Schweinezellen. Bei IP-Injektion von immunen Schweinezellen am 7. Tag (nach einer Thoraxbestrahlung am 3. Tag) war keine tumorhemmende Wirkung zu beobachten, d.h. der direkte Kontakt zwischen Schweine- und Tumorzelle ist wichtig. Schweinezellen, die gegen menschlichen Harnblasenkrebs immunisiert worden waren, hatten keinen Einfluss auf die Bildung von Tumoren in der Maeuselunge. (orig.)Additional details
