In vitro culture of soybean immature cotyledons and genetic transformation via biolistic system

Description

40 soybean (Glycine max (L.) Merr.) cultivars were collected to study in vitro culture of immature cotyledons. Regenerated plants of 3 cultivars were obtained and one cultivar was used to study the effect of irradiation treatment on stable genetic transformation rate. O, 5, 10, 20 Gy were applied before or after bombardment with Bio-Rad system. It was found that 5 Gy was the effective dose to raise the ratio of cotyledons with KmR calli. GUS enzyme activity showed that foreign genes had been integrated into genome of soybean callus. (authors)
