The influence of preparation parameters on the reliability of the thyroxine RIA kit RK-12

Neither significant differences (up to 50%) in the volume of the sample, of the antiserum or of the diluting buffer, nor changes in the duration or temperature of incubation, in the age of the radioactive tracer (125I-T4), and in the mode of the elimination of the supernatant after centrifugation (suction or decantation) influence the results, provided the alterations concern both the standard and the test sera. However, all these factors cause serious errors if the standard and test sera are prepared under different conditions. (L.E.)
