Radiation effects on testes. Pt. 15
Description
Effects of gamma-irradiation have been studied on NADP and NADP linked dehydrogenases of tests of rats after 720 and 2000 R surface exposures. After 720 R, activity of MDH (per mg protein and per 100 mg testes) declined between day 1 and 6 followed by a tendency of recovery from day 7 to 37. After a dose of 2000 R, minimum activity of MDH was observed on day 9; subsequently, it regenerated and became above normal value on days 25 and 33. Although the diphasic pattern of ME was similar to that of MDH at 2000 R, ME continued to decrease after 720 R over a period of 37 days. In contrast to MDH and ME, NADH2 diaphorase regenerated at faster rate after a transient loss during the first week. The synchrony observed between MDH and ME at 2000 R and association of hyperplasia of interstitial cells suggested that the pyruvate carboxylase pathway involving transhydrogenation plays an important role in lipogenesis of atrophied testes. Activity of LDH in whole testes was not influenced after 2000 R up to day 33, whereas it showed a significant increase on day 37 after 720 R. However, the analysis of different parts of testes suggested a significant loss of LDH in seminiferous tubulus and a stimulation in interstitial cells on day 30 after 720 R. Loss of LDH in seminiferous tubulus and loss of SDH in whole testes was an indication towards the loss of more matured germ cells of testes following irradiation. (orig.)

Die Wirkungen von Gammastrahlen wurden anhand von NAD- und NADP-gekoppelten Dehydrogenasen des Rattenhodens nach 720 und nach 2000 R OD untersucht. Nach 720 R verminderte sich die (per mg Protein und per 100 mg Testis gemessene) Aktivitaet der Malat-Dehydrogenase (MDH) zwischen dem 1. und 6. Tag, gefolgt von einer Tendenz zur Erholung vom 7. bis zum 37. Tag. Nach 2000 R wurde das Aktivitaetsminimum der Malat-Dehydrogenase am 9.Tag beobachtet; anschliessend regenerierte sie sich und ueberstieg am 25. und 33. Tag den Normalwert. Das Verhalten von Malatenzym (ME) nach 2000 R war zwar in aehnlicher Weise zweiphasisch wie das der Malat-Dehydrogenase, doch nach 720 R nahm es kontinuierlich 37 Tage lang ab. Anders als Malat-Dehydrogenase und Malatenzym regenerierte sich die NADH2-Diaphorase ziemlich schnell nach voruebergehenden Verlusten waehrend der ersten Woche. Das bei Malat-Dehydrogenase und Malatenzym beobachtete synchrone Verhalten nach 2000 R und das Hinzukommen einer Hyperplasie der Zwischenzellen fuehrt zu der Annahme, dass der Pyruvat-Decarboxylase-Reaktionsweg und der dabei erfolgende H2-Transport eine wichtige Rolle bei der Lipogenese atrophierter Hoden spielt. Die Aktivitaet der Lactat-Dehydrogenase (LDH) im ganzen Hoden blieb nach 2000 R bis zum 33. Tag unbeeinflusst, zeigte nach 720 R dagegen eine signifikante Zunahme am 37. Tag. Die Analyse verschiedener Hodenteile ergab einen signifikanten Verlust von Lactat-Dehydrogenase in den Samenroehrchen und eine Stimulierung der Zwischenzellen am 30. Tag nach 720 R. Die Verluste an Lactat-Dehydrogenase in den Samenroehrchen und die Abnahme der Sorbit-Dehydrogenase (SDH) im ganzen Hoden sind ein Hinweis auf den Verlust mehr ausgereifter Keimzellen des Hodens nach Bestrahlung.
