Dimercaptosuccinic acid-Tc99m: Preparation and biodistribution in rats

Description
Owing to the juxtaposition of 4 ligands (2 SH groups + 2 COOH groups), dimercaptosuccinic acid has a strong chelating capacity which suits it for technetium 99 m labelling. The study is carried out in 2 stages: preparation and stability of the dimercaptosuccinic acid - stannous chloride complex (DMSA-Sn); biodistribution of DMSA-Sn-Tc99m complex in rats as a function of the following parameters: pH, relative stannous chloride and dimercaptosuccinic acid concentrations, TcO4 volume added, injection time after labelling. The strong activity uptake obtained in rat kidneys represents a considerable step forward in the radioisotopic kidney examination and offers the prospect of clinical use
Abstract (French)
La juxtaposition de 4 ligands dans l'acide dimercaptosuccinique (2 groupements SH + 2 groupements COOH) lui confere un puissant pouvoir chelateur qui permet son marquage au Technetium 99m. L'etude comporte 2 parties. Preparation et stabilite du complexe acide dimercaptosuccinique-chlorure stanneux (DMSA-Sn); biodistribution chez le rat du complexe DMSA-Sn-Tc99m en fonction des parametres suivants: pH, concentrations relatives en chlorure stanneux et acide dimercaptosuccinique, volume de TcO4 ajoute, delai d'injection apres marquage. La forte captation de l'activite par les reins obtenue chez le rat constitue un progres appreciable dans l'examen radioisotopique du rein et permet de prevoir son utilisation cliniqueFiles
