Tissue distribution and excretion of 14C-labeled cinnamic aldehyde following acute subacute oral administration in male Fischer-344 rats

Carbon-14 labeled cinnamic aldehyde (CNMA-3-14C) was given orally (acute and subacute tests) to male rats at doses of 5, 50 and 500 mg/kg. Following a single oral dose, the radiolabeled CNMA distributed mainly to the GI tract, liver and kidney. Radiolabel excretion was primarily via urine; within 24 hr an avg. of 83.5% of the dose was recovered in the urine, and fecal excretion ranged from 3.2-5.1% at all dose levels. At all dose levels, some radiolabel distributed to the fat and was easily detectable in animals sacrificed 3 days after dosing. Less than 0.1% of radiolabel remained in the body of the rats 3 days after dosing. Following subacute (multiple) oral dosing, tissue distribution and excretion patterns were similar to those of rats observed after a single oral dose. Excretion via urine was rapid; in 24 hr, urinary excretion averaged 80.4%, 80.6% and 81.9% of the dose for 5, 50 and 500 mg/kg doses, respectively. Another 4.5-6.9% of the dose was recovered in the feces. As in the acute toxicity study, distribution was mainly to the rat liver, kidney and GI tract
