Effects of ionizing radiation on purinergic signaling modulation in rat brain nerve cells

Description

Purinergic signaling is composed of three modulatory components: a) source of extracellular nucleotides, b) specific receptor expression for these transmitter molecules and c) ectonucleotidase selection that dictate cell response gradually degradation extracellular nucleotides to nucleosides. ATP acts as a fast excitatory transmitter in the CNS. Postsynaptic actions of ATP are mediated by an extended family of purinergic, P2X receptors, widely expressed throughout the CNS. NTPDases hydrolyse extracellular ATP and ADP to AMP and are responsive for purinergfic termination. To investigate if ionizing irradiation could modulate CNS purinergic signalization we monitored activity of NTPDases and abundance of P2X7 receptor in synaptic plasma membranes after whole-body acute irradiation using low (0,5Gy) or therapeutic (2Gy) doses, 1h i 72h after irradiating juvenile (15-day old) and adult (90-day old) rats. Acute irradiation modulate purinergic system components investigated at the different ways in the rat development brain SPM and in the adult brain dependent of dose and time after irradiation

Abstract (Serbian)

Purinergicka signalizacija sadrzi tri modulatorne komponente: a) izvor vancelijskih nukleotida, b) specificne membranske receptore za ove transmiterske molekule i c) ektonukleotidaze koje diktiraju celijski odgovor postepenom degradacijom vancelijskih nukleotida do nukleozida. Adenozin trifosfat (ATP) je brzi ekscitatorni neurotransmiter u centralnom nervnom sistemu (CNS-u). Postsinapticko delovanje ATP-a posredovano je familijom specificnih purinergickih receptora, P2X, koji su siroko eksprimirani u CNS-u. Membranski enzimi nukleotid trifosfodifosfohidrolaze (NTPDaze) hidrolizuju vancelijske nukleotide, ATP i ADP do AMP i odgovorni su za terminaciju purinergicke signalizacije. Da bi ispitali da li jonizujuce zracenje moze da modulise purinergicku signalizaciju u CNS, pracena je promena aktivnosti NTPDaza i kolicina receptora P2X7 u plazma membranama nervnih zavrsetaka nakon akutnog ozracivanja celog tela malom (0,5Gy) ili teraeutskom (2Gy) dozom 1h i 72h nakon zracenja kod pacova u juvenilnom (15-dana stare zivotinje) i adultnom (90-dana stare zivotinje) uzrastu. Akutno zracenje celog tela modulise ispitivane komponente purinergickog sistema u mozgu pacova i nacin modulacije zavisi od stepena razvica CNS-a, primenjene doze zracenja i vremena nakon ozracivanja
