Investigations of the metabolic behaviour of lespenephryl with the aid of radiopharmaceutical methods

Some phases of the effect mechanisms of the two existing pharmaceutical forms of LESPENEPHRYL (R), were examined applying LESPENEPHRYL (R) labelled with tritium and non-labelled LESPENEPHRYL (R) in order to find out its points of affect with the help of the isotope method. The tests were carried out at rabbits and man, determining 1) the intestinal resorption and the excretion in the urin quantitatively 2) the distribution in the organism after intravenous injection 3) the effects on the renal function with the help of the isotopic-glomerular and tubular clearance-measurement 4) the kinetic distribution of the substance in animal organism in order to explain the metabolic behaviour better. Before starting the tests, problems of labelling, cleansing, and durability of tritium-labelled LESPENEPHRYL (R) were dealt with. (orig./MG)

Von den zwei bestehenden pharmazeutischen Anwendungsformen des LESPENEPHRYL (R) wurden einige Phasen des Wirkungsmechanismus, sei es unter Verwendung des - mit Tritium markierten LESPENEPHRYL (R), oder - von nicht markiertem LESPENEPHRYL (R) untersucht, um mit Hilfe des Isotopenverfahrens seine Angriffspunkte zu bestimmen. Die Versuche wurden am Kaninchen und beim Menschen durchgefuehrt und dabei 1. die Darmresorption sowie die Ausscheidung im Urin und in den Faeces quantitativ, 2. die Verteilung im Organismus nach intravenoeser Injektion, 3. die Wirkung auf die Nierenfunktion mit Hilfe der isotopisch-glomerulaeren und tubulaeren Clearance-Messung, 4. die kinetische Verteilung der Substanz im Tierorganismus, bestimmt, um so das Stoffwechselverhalten naeher zu klaeren. Vor Untersuchungsbeginn wurden Fragen der Markierung, Reinigung und Haltbarkeit des mit Tritium markierten LESPENEPHRYL (R) untersucht. (orig./MG)Additional details
