[en] The adsorptive behaviour of folic acid on the surface of a mercury electrode was investigated and the surface rate constant of electro-chemical reaction was determined (ks=44 s-1, 25 deg C). Adsorptive stripping voltammetry was applied to a flow system for determination of ultratrace amounts of folic acid. The effects of various parameters, for e.g., flow-rate, flow time of solution and accumulation potential, on peak currents are discussed. Interferences from the test solution can be eliminated by the technique of medium exchange. The peak current is proportional to the concentration of folic acid in the range of 10-8-10-11 mol l-1 with correlation coefficient of 0.998. The relative standard deviation is 2.5% at a concentration of 2.8x10-8 mol l-1 (7 determination). The detection limit of a.c. adsorptive stripping voltammetry is as low as 2x10-12 mol l-1 (10-min accumulation). (author). 10 refs.; 7 figs
[en] In Ethiopia, higher proportions of pregnant women are anemic. Despite the efforts to reduce iron deficiency anemia during pregnancy, only few women took an iron supplement as recommended. Thus, this study aimed to assess compliance with iron-folate supplement and associated factors among antenatal care attendant mothers in Misha district, South Ethiopia. Method. Community based cross-sectional study supported with in-depth interview was conducted from March 1 to March 30, 2015. The sample size was determined using single population proportion to 303. Simple random sampling technique was used to select the study participants. Bivariate and multivariable logistic regression analyses were employed to identify factors associated with compliance to iron-folate supplement. Results. The compliance rate was found to be 39.2%. Mothers knowledge of anemia (AOR = 4.451, 95% CI = (2.027,9.777)), knowledge of iron-folate supplement (AOR = 3.509, 95% CI = (1.1442,8.537)), and counseling on iron-folate supplement (AOR = 4.093, 95% CI = (2.2002,8.368)) were significantly associated with compliance to iron-folate supplement. Conclusions. Compliance rate of iron-folate supplementation during pregnancy remains very low. This study showed that providing women with clear instructions about iron-folate tablet intake and educating them on the health benefits of the iron-folate tablets can increase compliance with iron-folate supplementation.
[en] The diagnostic value of the Becton Dickinson Radioassay Kit (125I) for the assay of red cell folate has been investigated. The assay was acceptable with regards to precision but was non-linear with changing packed cell volume. Sensitivity of the assay was satisfactory, with 24 of 25 folate deficient patients giving red cell folate values which fell below the reference range. Specificity of the assay in the detection of folate deficiency was less satisfactory. As with microbiological assays, a considerable proportion of vitamin B12 deficient patients had low red cell folate values. In addition, low concentrations were found in 12% of patients who were unlikely to be deficient in either vitamin B12 or folic acid. (author)
[en] The aim of the present research was to formulate and characterize radioiodinated folic acid-chitosan conjugated thymoquinone nanoparticles (FATQCSNPs) and to increase targeting ability on ovarian cancer cell. The dose of drug-loading into the FATQCSNPs and the amount of folic acid on the FATQCSNPs surface were determined as a 20.0 ± 1% and 46.0 ± 0.5%, respectively. Cell viabilities (%) determined on SKOV-3 and Caco-2 cells for 48 h. TQ, TQCS and FATQCS were very cytotoxic with lower IC50 values on both cell lines. At specific-activity-dependent incorporation study, the incorporation efficiencies of 131I-FATQCSNPs was higher than that of 131I-TQ on SKOV3 cell lines. (author)
[en] ClGa 5,10,15,20-Tetrakis (4-bromophenyl) porphyrinato (ClGaTBrPP) and its metal free derivative were successfully synthesized and incorporated into Pluronic F127 polymeric micelles that has been conjugated to folic acid (FA) to form ClGaTBrPP-F127-FA (or H2TBrPP-F127-FA). For comparison, ClGaTBrPP-F127 and H2TBrPP-F127 (without FA) were also formed. The singlet oxygen quantum yield for ClGaTBrPP-F127-FA was higher (ΦΔ = 0.44) than that of H2TBrPP-F127-FA (ΦΔ = 0.37), due to the heavy atom effect of Ga in the former which encourages intersystem crossing to the triplet state. The same applies to ClGaTBrPP-F127 (ΦΔ = 0.47) and H2TBrPP-F127 (ΦΔ = 0.41). Thus, ΦΔ values decreased in the presence of FA, but still high enough for practical application of the nanodrug system. The binding constants Kb were determined to be 1.08 × 104 M−1, 2.51 × 105 M−1, 1.52 × 103 and 2.68 × 103 for H2TBrPP+F127-FA, ClGaTBrPP+F127-FA, ClGaTBrPP-F127 and H2TBrPP-F127, respectively. The Kp values were determined in biphasic octanol and water system.
[en] Folic acid plays a key role in nucleic acid biosynthesis, essential for normal cell proliferation and function. Localized folate deficiencies may be related to changes in cytology associated with cancer development; analogs of folic acid, such as methotrexate, are potent chemotherapeutic agents and are widely used either alone or in combination therapy for many types of cancer. In this report we describe the synthesis of a tetra-deuterated folic acid from perdeuterated toluene. The primary intermediate, N-(4-amino [2,3,5,62H4]benzolyl)-L-glutamic acid diethyl ester was coupled to N(2')-acetyl-6-formylpterin to create [2',3',5',6'-2H4]folic acid. A similar scheme can be used for the preparation of [1',2',3',4',5',6'-13C6] folic acid [13C6] ring labeled toluene. (author)
