AbstractAbstract
[en] The authors describe a case of bilebronchial fistula caused by a gun shot, that traversed the liver and right lung. The scintigraphic images with sup(99m)Tc-DISIDA showed the fistulous path sharply defined, thus evidencing the specificity of the method. The mechanism of cough, provoked during scintigraphy, improved the quality of the images and additionally increased the sensitivity of the method. The revision of literature and the report of this case confirm this procedure as preferential for the investigation of biliary fistulae. (author)

AbstractAbstract
[en] Various TSH RIA kit components were prepared. Conditions for 125I labelling of h-TSH were optimized by diminishing the amount of chloramine-T, extending reaction time and lowering reaction temperature. Yield, specific activity, and immunological activity could be maintained moderately under such mild reaction conditions. The mixture of polyethyleneglycol(PEG) and second antibody worked effectively as a B/F separation agent. Even though the mixture was made with more diluted PEG and second antibody than those of using the sole component separately, the time required for the B/F separation was shorter in case of using the mixture. The sequential saturation technique was efficient than those of applying ordinary equilibrium saturation technique in assay sensitivity and assay precision points of view. (Author)

[en] An unpredictable and unbalanced response to a stimulus like TSH is indeed a striking characteristic of the follicles of the simple goiter. Since it is known that the first step for TSH action on its target cell is binding to specific TSH plasma membrane receptors, the binding of 125ITSH to these receptors was studied in normal and ''cold'' hyperplastic thyroid fragments obtained at surgery. Through the Scatchard analysis it was verified that there are no differences with regard to the binding capacity of TSH receptors between normal and hyperplastic tissues. On the other hand, a significant decrease of the dissociation constant (Kd) was observed in hyperplastic tissue indicating higher affinity for TSH binding. It is known that intracellular iodine content can interfere with the TSH induced modifications on the thyroid folicular cells. It is supposed that this is mediated by interference on TSH binding to its receptor and/or activation of adenylate cyclase. Due to impaired organification capacity of ''cold'' tissue it is assumed that these cells present decreased intracellular iodine content. Therefore it is proposed that alterations of TSH binding to its receptors detected in the present investigation are consequent of the low iodine content of the hyperplastic folicular cell. (author)

Acredita-se que o desarranjo metabolico celular existente no bocio simples possa ser consequencia de um desequilibrio das acoes do TSH sobre a glandula tiroide. Ja que a ligacao do TSH ao seu receptor e a primeira etapa na genese de seus efeitos biologicos a nivel da celula alvo, objetivou-se na presente investigacao caracterizar a ligacao de 125ITSH ao seu receptor em fragmentos de tiroides humanas de pacientes portadores de nodulo ''frio'' e anatomopatologico de hiperplasia folicular benigna. Os tecidos foram obtidos no ato cirurgico e imediatamente armazenados em nitrogenio liquido. Apos determinacao das condicoes otimas de incubacao (duracao, pH do meio e concentracao proteica) e empregando-se a analise de Scatchard, verificou-se nao haver diferencas significativas quando a capacidade de ligacao dos receptores de TSH, quando comparada a do tecido normal paranodular. Por outro lado, observou-se nitida diminuicao da constante de dissociacao desses receptores, indicando maior afinidade pelo TSH. Sabe-se que o iodo intracelular pode interferir nas acoes do TSH sobre a celula folicular tiroidiana. Supoe-se que isto seja decorrente de um efeito de ligacao do TSH ao seu receptor. Devido a sua incapacidade de organificacao do iodo, os nodulos ''frios'' apresentam diminuicao do conteudo de iodo intracelular. Assim, acredita-se que as alteracoes encontradas no presente estudo sejam consequencia do baixo conteudo de iodo da celula folicular hiperplasica. (autor)Original Title

AbstractAbstract
[en] Gene-sequencing studies have shown that the glucagon precursor contains two additional glucagon-like sequences, the so-called glucagon-like peptides 1 and 2 (GLP-1 and GLP-2). The authors developed radio-immunoassays against synthetic peptides corresponding to these sequences. Antisera were raised in rabbits after carbodiimide conjugation of peptides to BSA. The selected antisera showed neither mutual cross-reactivity nor cross-reacted with any other peptide of the glucagon family. Trypsin digestion experiments showed that both antisera were directed against the C-terminus of the antigen peptides. I125 labelling was performed with chloramine T. and separation with plasma-coated charcoal. The detection limit was 7 and 25 pmol/l for GLP-1 and GLP-2. Accurate measurement of both peptides in plasma required extraction. Optimum recovery was obtained with ethanol at 75% (final concentration). The concentrations measured in fasting plasma from 10 normal subjects were 107 +- 7 pmol/l and 151 +-14 pmol/l for GLP-1 and GLP-2, respectively. After a mixed meal the concentrations rose slowly for 2 h reaching 145 +- 13 and 225 +- 15 pmol/l. (author)
