AbstractAbstract
[en] The purpose of this study was to design and evaluate a novel platform for labelling of Affibody molecules, enabling both recombinant and synthetic production and site-specific labelling with 99mTc or trivalent radiometals. The HER2-specific Affibody molecule PEP05352 was made by peptide synthesis. The chelator sequence SECG (serine-glutamic acid-cysteine-glycine) was anchored on the C-terminal to allow 99mTc labelling. The cysteine can alternatively serve as a conjugation site of the chelator DOTA for indium labelling. The resulting 99mTc- and 111In-labelled Affibody molecules were evaluated both in vitro and in vivo. Both conjugates retained their capacity to bind to HER2 receptors in vitro and in vivo. The tumour to blood ratio in LS174T xenografts was 30 at 4 h post-injection for both conjugates. Biodistribution data showed that the 99mTc-labelled Affibody molecule had a fourfold lower kidney accumulation compared with the 111In-labelled Affibody molecule while the accumulation in other organs was similar. Gamma camera imaging of the conjugates could clearly visualise the tumours 4 h after injection. Incorporation of the C-terminal SECG sequence in Affibody molecules provides a general multifunctional platform for site-specific labelling with different nuclides (technetium, indium, gallium, cobalt or yttrium) and for a flexible production (chemical synthesis or recombinant). [en] Isomers for elements far a way from line of stability are a new field for research to produce artificial isomers that can store considerable amount of energy in small amount of mass without the dangerous hazards on life and environment. These isomers could have very short life time or very long one 10th and 100th of years. It will be possible to get the stored energy by stimulated emission . The purpose of this work to use a theorical model based on Bcs method to find out the possible isomers in the mass reg on 178-192 even -even isotopes so that experimentalists can concentrate their research on these predicted isomers.

[en] This paper is not a general review of research progress in nuclear isomerism. This year brings the 90th anniversary of the birth of I.V. Kurchatov and the 50th anniversary of the founding of the Institute which bears his name. In this special year, the authors recall the history of the discovery of the effect and discuss the present status of the problem. They attempt to show that nuclear isomerism is still a 'live' and interesting field of nuclear physics, open for research and practical applications. Two broad classes of isomeric states in nuclei are discussed: spin isomers and spontaneously fissionable isomers. 44 refs., 5 figs
