AbstractAbstract
[en] A radiochemical ligand binding assay for methotrexate is provided. A binder factor comprising a partially purified dihydrofolic acid reductase preparation is employed. The binder factor is conveniently prepared by homogenizing a factor containing animal organ such as liver, and extracting with isotonic saline and ammonium sulfate. A binder cofactor, NADPH2, is also employed in the binding reaction. The procedure contemplates both direct and sequential assay techniques, and it is not interfered with by vast excesses of many natural folate derivatives. 12 claims, 6 drawing figures

AbstractAbstract
[en] This invention concerns the vessels used in radiation counting to determine radioactivity levels. These vessels prove to be particularly useful in analyses of the kind where a radioactive element or compound is separated into two phases and the radioactivity of one phase is determined. Such a vessel used in the counting of radiation includes an organic plastic substance tube appreciably cylindrical in shape whose upper end is open whilst the lower end is closed and integral with it, and an anti-radiation shield in metal or in metal reinforced plastic located at the lower end of the tube and extending along the wall of the tube up to a given height. The vessel contains a reaction area of 1 to 10 ml for holding fluid reagents

[en] Serum protein comprising specific binding proteins such as antibodies, B12 intrinsic factor, thyroxin binding globulin and the like may be copolymerized with globulin constituents of serum by the action of ethylchloroformate to form readily packed insoluble precipitates which, following purification as by washing, are eminently suited for employment as competitive binding protein absorbents in radioassay procedures. 10 claims, no drawings

AbstractAbstract
[en] A radioimmunoassay for a breast cancer associated antigen identified with two monoclonal antibodies (115D8/DF3) has been recently developed. The authors studied the behaviour of this new marker before using it routinely. From data of 107 normal subjects they chose a positive/negative cut-off of 31 u/ml (mean+3 SDs). High positivity rates and very high levels were found in advanced breast cancer; in primary breast cancer the CA15/3 serum levals were related to both tumor size and lymphnodal status. Low positivity rates were found in other advanced malignancies (lung, large bowel, esophagus, head and neck); however, high values were found in ovarian cancer patients. A high number of positive cases, but not very high levels, were found in a group of patients with mild or severe hepatic disease. Finally, the antigen was found in the cytosol of both breast carcinoma and, at lower levels, of normal breast tissue

AbstractAbstract
[en] This invention falls within the area of quantitative or semi-quantitative analysis by gamma spectrometry and particularly refers to a device for bringing the samples into the counting position. The purpose of this invention is precisely to provide an automatic apparatus specifically adapted to the analysis of hard gamma radiations. To this effect, the invention relates to a gamma spectrometry analytical device comprising a lead containment, a detector of which the sensitive part is located inside the containment and additionally comprising a transfer system for bringing the analyzed samples in succession to a counting position inside the containment above the detector. A feed compartment enables the samples to be brought in turn one by one on to the transfer system through a duct connecting the compartment to the transfer system. Sequential systems for the coordinated forward feed of the samples in the compartment and the transfer system complete this device

AbstractAbstract
[en] The authors used an immunoradiometric assay for serum antigen CA-125 in a survey of 92 patients affected by epithelial ovarian carcinoma. With this assay, positive values for cut-off of 35 U/ml were found in 85% of patients with active disease. Longitudinal levels of CA-125 were in agreement with clinical course in 23 out of 26 patients monitored serially; a discordant increase occurred in the remaining 3 patients. Levels of CA-125 may aid in monitoring the disease in patients with epithelial ovarian cancer and in certain instances they may reveal recurrence several months before the corresponding clinical diagnosis
